Sound set up not a VDJ issue.
Using VMS2 with EV powered speakers. ZXA1 8 inch.
Setting levels on VMS2 by adjusting master to approx 5.
Channel faders all the way up. Then adjust gain so the level just taps
into amber. Once this is completed I usually have the speaker volume
set to 1.
I see some people are setting the speaker volume all the way to max,
and some have said that this is the way it should be.
Just trying to get your thoughts. Is it best to have speaker volume
full on or set to a different level?

In my opinion it depends on the kit you're using...

With cheaper source kit, things like hiss and hum can make it impractical to crank up the amps or powered speakers to max.

It becomes easier to do when your source kit has a nice clean output which doesn't result in unpleasant noise when the gains are raised.
