Currently on Windows I can set my home folder location to a separate partition (for example D drive) which is the same as my external SSD and handy for syncing.
If I were to use my SSD on a Mac, would it be possible for VDJ to read the home folder directly from the SSD or does it still have to be in /Library/Application Support/VirtualDJ?

No, it's not possible to change on MacOS.
In theory you should be able to create a symlink from the current folder to a folder on your other drive, but I have never managed to succeed in making it.
Not to mention that MACOS these days tends to freak out with such approaches :P
